I don't think so. The slot 12 ROM is on the Plus 1 and the original plus 1 was designed for a 2764 (8K) and (correct me if I'm wrong..Dave) the Pres Plus 1 for a 27128 (16K) so either a 27256 or a 28C256 will work ok in that socket when using the appropriate AP6 image.
Edit: the 2764 uses addresses up to A12 and the 27128, A13 which are on the same pins on the 2764, 27128, 27256 and 28C256. It's only on the AP6 where A14 is used to split the 28C256 into two banks.
